Ultra Rapid Self-Contained Biological Indicator for Steam Sterilization

Terragene® Bionova® BT224 is a self-contained biological indicator designed to monitor steam sterilization processes with an ultra-rapid fluorescence readout in just 20 minutes. It contains Geobacillus stearothermophilus (ATCC® 7953) spores at a population of 10⁶ per vial.

Rapid Fluorescence Readout

After the sterilization cycle, activate the indicator and place it in a compatible Terragene® Bionova® Auto-Reader Incubator. Incubate for up to 20 minutes at 60 ± 2 °C.

• No fluorescence after 20 minutes: Negative result, indicating no detected biological growth.
• Fluorescence detected: Positive result, indicating a sterilization process failure.
• A non-sterilized indicator from the same batch should be used as a positive control.

Optional Visual Confirmation

A visual pH color change can be evaluated after 48 hours. If sterilization is unsuccessful, the culture medium may change from purple to greenish and then yellow, indicating the presence of viable spores. If sterilization is successful, the medium remains purple.

A 7-day visual readout may be performed for initial validation of the 20-minute fluorescence result but is not intended for routine use. If performed, a humidified environment is required to prevent the culture medium from drying.
